Extraction of a steroid 1,2 dehydrogenase by sonication of Arthrobacter simplex cells

Abstract

Extraction of a steroid 1,2 dehydrogenase was achieved by sonication after stabilization by the addition of monohydrated sodium glutamate (1.5 M). A specific first order rate of enzyme release of 0.035 min-1 was obtained. Studies were carried out on the following sonication parameters: power of sonication, cell concentration and container volume. A total recirculating system appears to be an advantageous approach to process larger volumes.
