Resumo
A new HPLC method for separation and quantitation of 19 protein amino acids is described. No pre- or post-column derivatization is necessary. Separation is achieved on a mixed reversed-phase ion-exchange column with continuous monitoring of the column eluent by an Evaporative Light-Scattering Detection device (ELSD). Separation of 19 protein amino acids is obtained in a single 60 minutes run. A linear log-log relationship between integrated peak area vs. analyte mass is observed in a range of 50 to 1.330 ng with a detection limit of 10-20 ng and mean recoveries of 94%. Quantitative determinations can be made with a single calibration function for all the amino acids.
